– Résumé :

Le palmier dattier (Phoenix dactylifera L.) est une culture fruitière d'une grande importance économique, nutritionnelle et culturelle dans les régions arides et semi-arides du Moyen-Orient et d'Afrique du Nord. Nous présentons ici un génome de référence sans lacune, assemblé de télomère à télomère, pour la variété ʿajwah, ainsi que dix-neuf assemblages génomiques supplémentaires de haute qualité — dix-huit femelles [dont le barnī d'al-ʿUlā] et un mâle. Ces données fournissent des assemblages chromosomiques révisés et corrigés, validés par cytogénétique, par Hi-C et par cartographie optique. La numérotation des chromosomes a été revue sur la base de leur longueur moyenne dans l'ensemble des génomes séquencés. La région de détermination du sexe (SDR) a été localisée par inférence sur le chromosome 14, où elle s'étend sur environ 15,2 Mb. Les régions organisatrices nucléolaires (NOR) ont été localisées sur le chromosome 10, où un vaste locus d'ADNr 45S présentait des motifs répétitifs uniques au sein des espaceurs, comprenant des séquences apparentées à des transposons. Chez certaines accessions, une seconde NOR a été mise en évidence sur le chromosome sexuel femelle. Cet ensemble d'assemblages génomiques du palmier dattier, structuré autour du génome de référence de la variété ʿajwah, constitue une ressource fondamentale pour le développement de stratégies d'amélioration variétale visant à renforcer la résilience génétique et la productivité du palmier dattier.

– Abstract

Phoenix dactylifera L. is an economically, nutritionally, and culturally important fruit crop in the arid and semi-arid regions of the Middle East and North Africa. Here, we present a gap-free, telomere-to-telomere reference genome of the variety Ajwa, along with 19 additional high-quality assemblies (18 female [including the barnī of al-ʿUlā] and 1 male). These assemblies reveal updated, corrected chromosome assemblies validated through cytogenetics, Hi-C and optical mapping. Chromosome numbering was revised based on average lengths across all sequenced genomes. The Sex Determination Region (SDR) was inferred on chromosome 14, spanning approximately 15.2 Mb. Nucleolar organizing regions (NORs) were localized on chromosome 10, where a large 45S rDNA locus displayed unique repeat spacer motifs containing transposon-like sequences. In some accessions, a second NOR was identified on the female sex chromosome. This collection of date palm assemblies, anchored by the Ajwa reference genome, provides a fundamental resource for advancing breeding strategies aimed at enhancing the genetic resilience and productivity of date palm.

L'ensemble des échantillons de plantes représentant une partie des connaissances ethnobotaniques des habitants de l'oasis d'al-ʿUlā (désert du Ḥijāz, Arabie saoudite) est déposé dans les collections de l'Herbier national (Ethnobotanique PAT) au Muséum national d'histoire naturelle.

Ces échantillons ont été exploités dans le cadre des travaux de recherche du projet al-ʿUlā Anthropological Survey, combinant de façon interdisciplinaire des approches anthropologiques, ethnoécologique et ethnobotanique.

The complete set of plant specimens representing a part of the ethnobotany knowledge of the inhabitants of the oasis of al-ʿUlā (Ḥijāz Desert, Saudi Arabia) has been deposited in the collections of the French National Herbarium (Ethnobotany, PAT) at the Muséum national d'histoire naturelle (MNHN).

These specimens have been used in the research project al-ʿUlā Anthropological Survey, combining anthropological, ethnoecological and ethnobotanical approaches within an interdisciplinary framework.

The complete set of date palm (Phoenix dactylifera L.) specimens representing the agrobiodiversity—that is, the diversity of cultivated forms—of the oasis of al-ʿUlā (Ḥijāz Desert, Saudi Arabia) has been deposited in the collections of the French National Herbarium (Ethnobotany, PAT) at the Muséum national d'histoire naturelle (MNHN).
